Selling on Ebay is no different than any other sale. The most important thing is to be confident that you have answered all the questions the customer might have. If someone takes interest, that's half the sale. Ask for the order. Treat your questions as if they were a closed deal before you get off the phone. Ask questions such as "Will that be your credit card or paypal?" Open ended closing questions are important. "Do you want it shipping overnight or ground?" This gets the customer to re-inforce that you have a deal.
